Also a disclaimer about the seed: Because I had to film things specifically for this tutorial instead of editing the video after the fact, the footage in this tutorial is not of an official run. The over world and nether are different worlds that I used for demonstration purposes. The overworld seed is: 7513025135232960962 @cord(x=776, z=136) and the nether seed is: 9179612639808263344 @ cord(x=512,z=436 (In the nether))

Details:
========= Reseting Runs =========
Minecraft version 1.16.1 has the potential to be the most optimal and fast way to speedrun the any% random seed 1.9+ category. However, it is extremely RNG based and will require many resets before a successful run. If you are interested in speedrunning, but would like to start of with a more consistent version, I would recommend speedrunning version 1.14.4. I made a tutorial for that version as well which can be found in the links above.
Don't Play Ver 1.16.2. It's bad. (ender pearls harder to get)
========= Desert Temple =========
If you do find a desert temple you wan to grab:
Iron (For Bucket & Flint n Steel)
Diamonds (You can make a diamond tool to help speed things up. Even a diamond shovel can help)
Gold (For trading with Piglins)
3 String (For bow)
Sand (For misc building and to save time so you don't have to mine blocks)
Golden Apple(For situations when your low on health. e.g killing blaze in the nether)
Saddle (In case you find a horse you can use it to get to stronghold faster)
========= Iron Golem ==========
The iron golem can drop 35 iron. If it drops only 3, you need to get 1 more iron from either a desert temple or finding some in a cave or structure chest somewhere.
========= Nether Prep ==========
Make sure that you have some extra cobble and sticks to build an extra pickaxe in the nether. Because you need to mine alot of gold ore, and might have to mine netherack to traverse around, you'll probably need to build another pickaxe in the nether. Also that means to make sure and bring your crafting table as well.
========= The Nether ===========
Okay this is the scary part of 1.16 speedruns. It is significantly harder to find a nether fortress compared to other versions. Also if you spawn in certain bioms, like the soul sand valley, it can be very difficult to get around and find a fortress. It's okay to spend alot of time looking for the fortress. This is where most runs get reset.
========= Piglins Bartering =========
Piglins have a 4.73% chance of dropping 48 ender pearls. So on average it will take about 3040 ingots to get 12 or more ender pearls. This is approximately 5 stacks of gold nuggets. You can try to trade with them after you get about 1020 ingots and if you don't get the pearls, go back to mining again. If the Piglins don't drop the pearls after 40 ingots you might want to reset.
========= Arrows ==========
The number of arrows you need will just depend on how good you are with a bow. I highly recommend practicing the ender dragon fight in a creative world. Then you can see on average how many arrows it takes you to shoot down all the crystals and go from there.
========= Finding the Stronghold ==========
This part is pretty self explanatory but if you want to see how long I usually run for before throwing a pearl, you can check out my 1.14.4 speedrun in the links above. (I'm actually pretty bad at that part lol)
========== Ender Dragon Fight =============
The dragon will perch and do it's breath attack if the player is within 150 blocks of the end fountain. It will do the breath attack up to 4 times or until it takes 25 hearts of damage. Once it takes 25 hearts of damage it will fly off. The dragon has 100 hearts of health so after you do a little less than 25% of it's health with a sword, you can use a bed to deal alot of extra damage before it flies off.
